Molecular Phylogeny of Six Species Belonging to Cactaceae and Aizoaceae Families using Start Codon Targeted (SCoT) Polymorphism and in silico matK Barcode Analysis
Abstract
Halophytes have been used throughout the centuries for their therapeutic properties, while there is a recent increase in global demand for herbal medicines and food supplements. The Aizoaceae and Cactaceae are two huge plant groups belonging to the halophytes that involve a massive number of species. But, there are no sufficient studies that manipulate the molecular phylogeny of these two families, and they are still in a disconcerting status. Our study contributes fundamental information for the phylogenetic analysis of the Aizoaceae and Cactaceae families. Ten SCoT primers were used to study the genetic diversity of six halophyte species; F. glaucescens, F. herrerae, F. pottsii, G. linguiforme, A. cordifolia var. variegata, and C. edulis. Based on the analysis of markers indices the primers SCoT-6, SCoT-7 and SCoT-8 have a significant discrimination power and are suitable for the detection of the genetic diversity in the studied species and it may be concluded on the basis of this study that the genetic diversity analysis with SCoT markers was more effective. Besides, our results showed consistency with the previous studies for the classification and clustering of halophytes species. The in silico analysis of the matK genes and proteins also manifests a similar pattern to the SCoT markers analysis. In conclusion, the F. glaucescens, F. herrerae, and F. pottsii were clustered together as they are members of the Cactaceae family, while the G. linguiforme, A. cordifolia var. variegata, and C. edulis were clustered together as they are members of the Aizoaceae family
